Application

ADC internalization:

The antibody binds its target antigen on a tumor cell → ADC is internalized via endocytosis.

Lysosomal trafficking:

ADC enters the lysosome, which is rich in cathepsin B (a cysteine protease).

Enzyme cleavage:

Cathepsin B recognizes Val-Cit and cleaves the peptide between Cit and the PABC spacer.

Payload release:

The PABC self-immolative spacer undergoes 1,6-elimination after cleavage → releases the free payload in its active form.
