Application

 

Protection of the Amino Group: The Fmoc group in Fmoc-Lys(ivdde)-OH is used to protect the α-amino group of the amino acid. This ensures that during solid-phase peptide synthesis, the amino group does not undergo unintended condensation reactions with the carboxyl groups of other amino acids. This is an essential step in the Fmoc method of peptide synthesis, which was invented by American biochemist R. Bruce Merrifield in 1962.

 

Orthogonal Protection Strategy: The ivdde group serves as a protective group for the ε-amino group of lysine (Lys). Lysine is a diamino acid with a strongly basic ε-amino group that exhibits nucleophilic properties. During peptide synthesis, to avoid confusion between the ε-amino and α-amino groups, an orthogonal protection strategy is employed. The selective removal properties of the ivdde group make it particularly effective in this strategy.

 

Suitability for the Synthesis of Special Peptides: Due to the unique properties of the ivdde group, Fmoc-Lys(ivdde)-OH is particularly suitable for the synthesis of branched peptides, cyclic peptides, and peptides with modified side chain amino groups. This is because the ivdde group can be selectively removed under specific conditions while other protective groups, such as the Boc group, remain stable.

 

Practicality of the Synthesis Method: Studies on the terminal amino protection of four amino acids with different carbon chain lengths (Fmoc-Dap-OH, Fmoc-Dab-OH, Fmoc-Orn-OH, and Fmoc-Lys-OH) using the ivdde group have shown that the synthetic methods obtained are practical and suitable for large-scale preparation.
