Application

Protection of the Amino Group: The Fmoc group in Fmoc-Lys(ivdde)-OH is used to protect the α-amino group of the amino acid. This ensures that during solid-phase peptide synthesis, the amino group does not undergo unintended condensation reactions with the carboxyl groups of other amino acids. This is an essential step in the Fmoc method of peptide synthesis, which was invented by American biochemist R. Bruce Merrifield in 1962.

Orthogonal Protection Strategy: The ivdde group serves as a protective group for the ε-amino group of lysine (Lys). Lysine is a diamino acid with a strongly basic ε-amino group that exhibits nucleophilic properties. During peptide synthesis, to avoid confusion between the ε-amino and α-amino groups, an orthogonal protection strategy is employed.
