Application
Protection of the ε-Amino Group:
The dde group in Fmoc-Lys(dde) is specifically designed to protect the ε-amino group of lysine (Lys). Lysine is a diamino acid with a strongly basic ε-amino group that exhibits nucleophilic properties. In peptide synthesis, the protection of the ε-amino group is essential to prevent unwanted chemical reactions.
The dde group, due to its unique chemical properties, effectively protects the ε-amino group during peptide synthesis, ensuring the correct formation of the peptide chain.
Orthogonal Protection Strategy:
To prevent confusion and undesired reactions between the α-amino and ε-amino groups in peptide synthesis, an orthogonal protection strategy is often employed. The Fmoc group protects the α-amino group, while the dde group protects the ε-amino group, allowing for efficient orthogonal protection during the synthesis process.
Suitability for the Synthesis of Special Peptides:
Due to the unique properties of the dde group, Fmoc-Lys(dde) is particularly suitable for the synthesis of branched peptides, cyclic peptides, and peptides with modified side chain amino groups. This is because the dde group can be selectively removed under specific conditions, while other protective groups, such as the Boc group, remain stable.
Ease of Removal and Stability:
The dde group can be readily removed using 2% hydrazine/DMF solution, while remaining stable in TFA solutions. This property allows for convenient removal of the dde group after peptide synthesis, resulting in the desired target peptide.
Product Purity and Synthesis Efficiency:
The use of Fmoc-Lys(dde) as a raw material for peptide synthesis, due to its excellent orthogonal protection and selective removal characteristics, can effectively reduce side reactions, leading to improved product purity and synthesis efficiency.
