Application

For lysine, which contains two amino groups - the α-amino group and the ε-amino group - specific protection is necessary.

 

In Fmoc-Lys(Mmt), the Fmoc (9-fluorenylmethoxycarbonyl) group serves as a temporary protecting group for the α-amino group. It is stable under acidic conditions but readily removed under basic conditions, allowing for further chain elongation. On the other hand, the Mmt (4-methoxytrityl) group permanently protects the ε-amino group, preventing it from reacting during the standard peptide synthesis process.

 

Enhancing Synthesis Efficiency:The use of Fmoc-Lys(Mmt) in peptide synthesis allows for enhanced efficiency due to its clear protection strategy. The selective protection of the α- and ε-amino groups prevents interference between them during the synthesis, resulting in smoother reactions and higher yields.

 

Additionally, the ease of removing the Fmoc group under basic conditions facilitates the extension of the peptide chain, making the synthesis process more flexible and efficient.
