Application

 

Fmoc-Tyr(tBu)-Aib-OH is a key intermediate used in the synthesis of tirzepatide, a dual glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptor agonist used for the treatment of type 2 diabetes and weight management.

 

Facilitating Solid-Phase Peptide Synthesis (SPPS): The Fmoc group protects the amine of the amino acid during SPPS, allowing controlled and sequential addition of amino acids to form the tirzepatide peptide chain. After coupling, the Fmoc group can be removed selectively using a mild base, leaving the free amine ready for the next coupling step. The tBu group can later be removed under acidic conditions, revealing the free phenolic hydroxyl group of tyrosine for functional roles in the final peptide.

 

Ensuring Precise Functionalization: The tyrosine residue with a protected hydroxyl group is critical for subsequent modifications or interactions with other functional groups in the tirzepatide structure. This ensures that the final product has the correct functional properties.
