Application
1. Cathepsin B-Cleavable Linker Component
The Gly-Gly-Phe sequence is recognized and cleaved by lysosomal proteases, particularly cathepsin B, which is overexpressed in many tumor cells.
Upon internalization of the ADC into tumor cells, the peptide is enzymatically cleaved, triggering drug release inside the cell.
2. Final Structure in Linker-Payload Design
Unlike Fmoc-protected versions (used in synthesis), Gly-Gly-Phe-OH is the active, deprotected form used in the final assembly of the ADC linker-payload unit.
It is typically conjugated to:
PABC (para-aminobenzyloxycarbonyl) as a self-immolative spacer,
then to the cytotoxic payload (e.g., MMAE, DM1, etc.).
