Application

 

1. Process Simplification

Fmoc-α-Me-Leu-OH only carries one protecting group (Fmoc) on the amino group, with a free carboxyl group and no need for side chain protection (since Leu has no reactive side chain).

Dual-protected amino acids often require additional deprotection steps (e.g., Boc/Fmoc switching, side chain removal), making the synthesis more complex and lowering overall yield.

 

2. Compatibility with Standard SPPS

Fmoc-amino acids are the mainstream choice for solid-phase peptide synthesis (SPPS) since Fmoc can be removed under mild basic conditions, without affecting peptide bonds.

Dual-protected amino acids may introduce incompatibility in automated synthesis, leading to side reactions or material loss.

 

3. Advantage of α-Methylation

The α-methyl substitution in α-Me-Leu increases conformational rigidity of the peptide bond and enhances resistance to enzymatic degradation (improves stability and biological activity).

A single Fmoc protection is sufficient for peptide assembly, and no additional protection is needed to stabilize the structure.

 

4. Cost & Practicality
