Application
Efficient SPPS compatibility
Fmoc strategy is the most common in automated solid-phase peptide synthesis.
The tBu protecting group on Tyr is stable under basic Fmoc-removal conditions but can be cleanly removed under strong acid (TFA) at the end.
Prevention of side reactions
Tyr’s hydroxyl group can participate in undesired acylation or oxidation; tBu protection prevents this.
Enhanced peptide stability
The α-methyl substitution increases conformational rigidity, reduces enzymatic cleavage, and may alter the peptide’s secondary structure for improved biological activity.
Selective deprotection
You can remove Fmoc during elongation (base-labile).
Keep tBu intact until final global deprotection (acid-labile).
This orthogonality is ideal for SPPS.
